New Doctor of Medicine Proves Corosolic Acid's Potential as an Anti-obesity Treatment

Obesity is a condition of excessive fat accumulation that poses health risks. Obesity sufferers are increasing from all walks of life both in the world and in Indonesia. Due to the high risk of obesity, several obesity treatments have been carried out ranging from lifestyle adjustments, drugs to surgery. Anti-Aging Medicine is one of the efforts that can be done to prevent or inhibit these conditions. Corosolic Acid (CA) is a compound that has antioxidant activity that is widely used in several slimming drugs that can help overcome obesity. PipingRock is a standardized supplement that contains 0.6 mg of CA in 30 mg of extract. The pathways targeted by CA are oxidative stress and anti-inflammatory pathways. This study aims to prove corosolic acid (CA) can reduce body weight, Malondialdehyde (MDA) levels, hydroxymethylglutaryl co-enzyme A (HMG-CoA) reductase, and increase leptin levels and glucagon like peptide-1 (GLP-1) levels in male Wistar rats (Rattus norvegicus) with obesity.
This study is a true experimental study using a randomized pre and post test control group design, which uses male Wistar rats with obesity. Rats were divided into two groups, namely the control group (P0) and the treatment group (P1), each group consisting of 10 rats. Plasma and serum were taken before (pre-test) and after (post-test) treatment. The variables examined included weight loss, MDA levels, HMG-CoA reductase, leptin and GLP-1. These variables were examined by ELISA method. Descriptive statistical analysis and comparative statistical analysis were performed using independent sample t-test or its alternative with p>0.05 considered as significantly different.
CA administration of 10 mg/Kg BW for 6 weeks to male Wistar rats with obesity was proven to reduce body weight, MDA levels, and HMG-CoA reductase of the P1 group post-test significantly at p & lt; 0.001. In addition, CA administration was proven to increase leptin levels and GLP-1 levels in the P1 group post-test significantly at p & lt; 0.001. The decrease in body weight in male Wistar rats was due to loss of appetite in rats which was also evidenced by the increase in GLP-1 levels in the P1 group post-test and the remaining food given to the P0 group was less than the remaining food of the P1 group. The decrease in MDA levels is related to the high antioxidant properties of CA so that it can inhibit the formation of ROS in the body. In addition, CA was proven to reduce the level of HMG-CoA reductase which plays an important role in cholesterol synthesis. Furthermore, CA was shown to increase leptin levels by helping to reduce fat levels in the body so that leptin production can increase as evidenced by weight loss.
Based on the results of this study, CA was shown to reduce body weight, MDA levels, HMG-CoA reductase levels, and increase leptin and GLP-1 levels in obese male Wistar rats. CA is very potential as a good antiobesity agent, through the mechanism of ROS.
